La Facultad nace luego de un extenso proceso que inició en 1999 cuando la Institución era aún una Corporación Universitaria, con la creación del Departamento de Ciencias Básicas, unidad que tenía como propósito ofrecer los cursos del área básica -biociencias, biología, química y bioquímica-, a los programas de la Facultad de Salud; posteriormente, se organiza el Departamento de Matemáticas y Física, con el objetivo de ofrecer los cursos en las áreas de matemática, física y estadística a los programas de las Facultades de Ingeniería, Administración y Salud.

- PublicaciónAcceso abiertoFactores Productivos Asociados a la Presencia de Miopatía Pectoral Profunda y Pechuga de Madera en Pollo de Engorde en Municipios del Departamento de Santander, Colombia(Universidad de Santander, 21-10-21) Rincón-Uribe, Josimar Sneider; González-Gamboa, Nidia Fernanda; Solano Marcixgclia, Jorge-AlbertoLos avances genéticos y nutricionales de los últimos 50 años para mejorar la eficiencia de la industria avícola saturaron el modelo animal, generando problemas metabólicos y estructurales en los músculos de la pechuga cambiando sus características organolépticas, generando impactos económicos en la industria. En el siguiente estudio se busca evaluar los factores asociados a la ocurrencia de pechuga de madera (PM) y miopatía pectoral profunda (MPP) en municipios del departamento de Santander, Colombia. Se realizó el análisis de 896 lotes en 173 predios, para un total de 8´987.242 pollos de engorde de 5-7 semanas de edad destinados a sacrificio con un peso promedio de 2.800g. Los datos fueron analizados en “SPSS Statistics V25”, realizando pruebas de asociación por X2 de Pearson, razón de prevalencia, correlación de Spearman y Odds ratio para la evaluación de riesgo. El nivel de confianza usado en la investigación fue del 95%. La ocurrencia general en los municipios de Santander de 9.0% para MPP y 18.9% para PM. Se encontró asociación estadísticamente significativa entre el desarrollo de MPP y las categorías de raza, sexo, tipo, edad y municipio, de igual manera para PM se encontró asociación entre sexo, tipo y municipio. Las variables productivas mostraron una correlación significativa, pero de baja intensidad. Los factores de riesgo se asociaron a la raza RossAp, macho y tradicional para MPP y macho, tradicional para PM. Se concluye que la presencia de MPP y PM se encuentra asociados a factores productivos y variables de los lotes como sexo, tipo, raza y municipio. Variables como la pigmentación desempeñan un rol protector frente a la presentación de MPP y PM.
- PublicaciónAcceso abiertoEvaluación de la Capacidad de Depuración Biológica de un Filtro tipo Airlift para el Tratamiento de Aguas Usadas Durante el Proceso de Extracción de Aceite Empleando un Soporte a Partir de Cuesco de Palma a Escala de Laboratorio(21-12-15) Gelves-Gamboa, María Camila; Chacín Zambrano, Christian-Andrei; Rodríguez González, Leidy-GabrielaLa palma africana (Elaeis guineensis) es el cultivo de mayor crecimiento en Colombia ya que abastece la mayor parte del mercado nacional de aceites y grasas. Sin embargo, la extracción de aceite de palma produce grandes cantidades de residuos, entre estos se encuentra el cuesco, que se encarga de proteger a la almendra, y, los efluentes resultantes del proceso, los cuales poseen altas cantidades de grasas y aceites. Es por esto, que la utilización de los residuos agroindustriales es una buena alternativa para la generación de otros productos de mayor valor agregado o aplicación a procesos que disminuyen el impacto ambiental que genera la disposición y manejo de estos. En este trabajo de grado se evaluó la capacidad de depuración biológica de un filtro tipo airlift a escala de laboratorio sobre aguas residuales del proceso de extracción de aceite de palma de una empresa palmera. a partir de una matriz no convencional de cuescos de palma. Para esto, inicialmente se estableció la formación de la biopelícula en la matriz de cuescos de palma con los microorganismos contenidos en la muestra de agua residual de las lagunas de oxidación de la empresa. Posteriormente, se construyó un filtro tipo airlift a escala de laboratorio y se puso en funcionamiento en diferentes tiempos de retención hidráulicos (TRH) con aireación y sin aireación, donde se evaluaron parámetros físico - químicos como la DQO, SST, nitratos y sulfatos para determinar la eficiencia del proceso. Se evidenció que el TRH de 12 horas con aplicación de aire, presentó mejores resultados en cuanto a las características físico – químicas de la calidad de las aguas usadas en la extracción de aceite de palma. Los resultados de la investigación permitieron contribuir a la generación de nuevo conocimiento sobre el aprovechamiento de residuos agroindustriales.
- PublicaciónAcceso abiertoPrevalencia y Factores de Riesgo Zoonótico de Leishmania infantum en Albergues Caninos y Áreas Adyacentes Ubicados en el Área Metropolitana de Bucaramanga(22-06-30) Suarez-Becerra, Daniela; Vega-Prada, Carlos Manuel; Florez-Muñoz, Angel AlbertoLa leishmaniasis es una patología transferida por la picadura de insectos conocidos como flebótomos, producida por parásitos protozoos de la familia Leishmania (Kinetoplastida: Trypanosomatidae) que causan problemas cutáneos como llagas, nódulos, alopecia, úlceras, dermatosis y también problemas viscerales que generan emesis, epistaxis, esplenomegalia afectando órganos como hígado, bazo, medula ósea entre otros, afectan a caninos de zonas tropicales y subtropicales del primer y tercer mundo. Por esto es necesario realizar un estudio epidemiológico del parásito para determinar la seroprevalencia y los factores de riesgo de la leishmaniosis canina. En el presente estudio se encontró una seroprevalencia general de 4.3% (Bucaramanga, Piedecuesta, Girón y Floridablanca), determinando como Girón la zona más afectada por leishmaniosis canina, sin embargo, el valor de la prevalencia puede estar disminuido debido a que la prueba ELISA solo reconoce pacientes con anticuerpos. Se realizó una investigación de tipo descriptiva, donde se recolectaron 196 muestras de sangre canina, las cuales fueron analizadas para le determinación del Ag de Leishmania infantum por medio del Ingezim Test, por cada canino inspeccionado se aplicó una encuesta epidemiológica al propietario la cual se ingresaba en el software Epi Info™ versión 7.2.2.6. Los valores se procesarán estadísticamente utilizando la prueba de Ji – cuadrado de Pearson (X²) y Odds Ratio. Todos los datos se analizaron empleando el programa estadístico SPSS versión 21®. La seroprevalencia encontrada en el presente estudio es menor a la de otros estudios nacionales realizados en los últimos años.
- PublicaciónAcceso abiertoEvaluación del crecimiento in vitro de maracuyá amarilla (Passiflora edulis SIMS forma flavicarpa) a partir de segmentos nodales mediante la técnica de organogènesis(Bucaramanga, Universidad de Santander, 2019, 2009-06-26) Ortiz Rodríguez, Lizeth Paola; Chacín Zambrano, Christian-AndreiLos cultivos de maracuyá se han logrado beneficiar gracias a su gran demanda de exportación a nivel internacional. En Colombia los cultivos de maracuyá entre los años 2014-2018 han aumentado de manera significativa. En el departamento de Santander se han presentado factores como enfermedades causadas principalmente por Fusarium oxysporum y Fusarium solani. Llevando esto a la baja producción en los cultivos de maracuyá generando pérdidas hasta una 60% de la producción, con el fin de presentar una alternativa viable para la multiplicación de forma sana, libre de patógenos y genéticamente uniformes, este trabajo tiene el objetivo evaluar el crecimiento in vitro de maracuyá amarilla a partir de segmentos nodales mediante la técnica de organogénesis. Por esta razón se estableció un proceso de desinfección con diferentes tipos de compuestos y tiempo de exposición donde se evidenció que T4 presento bajos índices de contaminación, oxidación y explantes prósperos, de igual forma se estableció un medio de cultivo donde se estableció que a bajas concentraciones 0,2 mg/l de 6-BAP se logra generar la formación de brotes con un porcentaje de 97% y un promedio de número de hojas de 1,80 por segmento nodales, igual manera la aplicación de arginina en concentración de 30 mg/l genera un mayor largo de las hojas de 2,9 cm.
- PublicaciónAcceso abiertoEvaluación del antagonismo y sinergismo en aislados microbianos obtenidos de plukenetia volúbilis L para la conformación de un consorcio microbiano en condiciones in vitro.(Bucaramanga : Universidad de Santander, 2017, 2017-06-06) Moreno Riaño, Cindy Paola; Agualimpia Valderrama, Bayron-Enrique; Acevedo Isidro, Carlos-Augusto; Osma Castellanos, Wolfang.El propósito de este trabajo fue determinar la compatibilidad de un grupo de microorganismos que fueron aislados de diferentes órganos de la planta sacha inchi (Plukenetia volúbilis Linneo) que presentaron actividad PGPR, estudiados en fases anteriores del macroproyecto de aspecto institucional, con el fin de establecer combinaciones microbianas que potencialicen las actividades de interés como solubilizacion de fosfatos y producción de sustancias indolicas; para ello inicialmente se evaluó el antagonismo mediante ensayos de inhibición de crecimiento utilizando pruebas de botón en césped y plato dual.Los microorganismos que no presentaron antagonismo se les evaluó el sinergismo por la tasa de variación de su actividad mediante técnicas colorimétricas de salkowski y moliddovanadato fosfórico, y finalmente se seleccionaron los microorganismos correlacionando las pruebas de antagonismo y sinergismo mediante el análisis estadístico. Se logró determinar que el 81% de los microorganismos no presentaron antagonismo, destacándose la cepa bacteria TSEBT04-03 quien no inhibió el crecimiento en el enfrentamiento bacteriano y que el asilado microbiano TSEBR01-01 presentó mayor inhibición (36,4%) en los enfrentamientos y su frecuencia represento en más del 50% en los ensayos. Finalmente se pudo establecer que el 35,93% de los microorganismos fueron compatibles, al no presentar antagonismo y aumentar su actividad al ser evaluados en conjunto, encontrándose que los consorcios significativos en el aumento para ambas actividades de interés fueron los consorcios C2, C6, C21 Y C31 de las 32 combinaciones evaluadas. Este trabajo permitió el establecimiento de un consorcio conformado por tres (3) microorganismos, que presenta menor capacidad antagónica y mayor sinergismo para las actividades PGPR evaluadas.

CCSP 89-43 a partir de meristemos apicales mediante la técnica de organogénesis.(Bucaramanga : Universidad de Santander, 2017, 2017-11-21) Betancourt Guerrero, Jessica Marcela; Chacín Zambrano, Christian-AndreiEl cultivo in vitro de tejidos vegetales en la actualidad ha tenido grandes avances, siendo de gran importancia la incorporación de esta técnica en distintos cultivos, para lograr la multiplicación y conservación de diferentes especies. El objetivo del presente proyecto de investigación fue evaluar el cultivo in vitro de caña de azúcar (Saccharum officinarum) variedad CCSP 89-43 a partir de meristemos apicales mediante la técnica de organogénesis. Para el desarrollo de esta investigación se trazaron tres fases, la primer fase fue la selección del protocolo, en esta fase se trabajó con cuatro protocolos de desinfección, los cuales fueron evaluados de acuerdo con el porcentaje de explantes prósperos, explantes contaminados y explantes oxidados, la segunda fase fue el desarrollo del medio de establecimiento, para esta fase se formularon tres medios de cultivo (MEc1, MEc2, MEc3), y la tercera fase, fue la multiplicación, en la cual igual que en la segunda fase se formularon tres medios de cultivo (MMc1, MMc2, MMc3). De acuerdo con el análisis de varianza (Anova) se determinó que el mejor tratamiento de desinfección fue el número dos, el cual contenía sustancias como: Solución jabonosa en un 3% con un tiempo de 2 minutos, alcohol en un 70% con un tiempo de 2 minutos, hipoclorito de sodio al 5% con un tiempo de 15 minutos, y ácido cítrico al 3% con tiempo de 10 minutos. En lo que respecta al medio de establecimiento el mejor fue el MEc2 el cual contaba con sales del medio MS, la hormona 6-BAP (500 μ/L) y la hormona AG3 (100 μ/L), obteniendo una germinación en un tiempo de 18 días, en el medio de multiplicación el que obtuvo mejores resultados de acuerdo con explantes multiplicados y longitud de las hojas fue el MMc3, el cual contenía sales del medio MS, Glicina (1500 μ/L) Arginina (1500 μ/L) y 6- BAP (750 μ/L).
- PublicaciónAcceso abiertoEvaluación de dos productos naturales a base de extractos de cítricos como alternativa de reemplazo del triclosán en jabones antibacteriales(Bucaramanga : Universidad de Santander, 2017, 2017-12-01) García García, Ahidaly Yineth; Laguado, Jenny Adriana; Contreras Contreras, Sandra LorenaCurrently, the regulatory bodies of Good Manufacturing Practices (GMP), have increased the requirements to ensure the quality of products and as a priority are focusing largely on hand cleaning and disinfection programs, and in production areas plants, due to the risks associated with cross-contamination that may occur between different food products. In the present study, two natural products based on citrus extracts were evaluated as an alternative to the replacement of Triclosán used in antibacterial hand soaps. For this, two (2) Kelsey Maurer and Kirby-Bauer techniques were performed. In the first set 90 seconds as the optimal time of effectiveness of extract 2 to 0.5%, from the logarithmic reduction found of the values obtained; and in the second, sensitivity was observed in the triclosán, and resistance in the extracts according to the size of the inhibition, for the microorganisms B.cereus, E.coli and S. aureus, and when using the protocol of hand washing recommended by The World Health Organization Organization (WHO), for the quantification of the fungal and bacterial flora in the hands, it was found that the use of Extract 2 as active, in antibacterial soap, significantly reduced the logarithmic units of bacteria and fungi in comparison with the triclosán.
- PublicaciónAcceso abiertoEvaluación del crecimiento in vitro del frijol arbustivo (Phaseolus vulgaris) tolerante al estrés hídrico mediante la técnica de organogénesis.(Bucaramanga : Universidad de Santander, 2018, 2018-01-19) Angarita Guzmán, Karen; Chacín Zambrano, Christian-AndreiEl frijol arbustivo en Colombia es de gran importancia en el sector agrícola y comercial (Arias Restrepo , Rengijo Martínez, & Jaramillo Carmona, 2007), por su gran contenido de proteinas y vitaminas como base nutricional de la población. Sin embargo se ha visto afectada su producción por malas prácticas agrícolas trayendo problemas fitosanitarios del cultivo en el departamento de Santander (Arias Restrepo , Rengijo Martínez, & Jaramillo Carmona, 2007). El objetivo de este trabajo de investigación fue establecer la propagación in vitro de Phaseolus vulgaris tolerante al estrés hídrico mediante la técnica de organogénesis, por esta razón se seleccionó un protocolo de desinfección a partir de tres tratamientos con diferentes tipos de desinfectantes a distintas concentraciones y tiempos de exposición de los embriones. Los resultados mostraron que el tratamiento T3 tuvo una mejor actividad de remoción de impurezas y eliminación de microorganismos (Solución jabonosa, Alcohol al 70%, Hipoclorito de sodio al 5% por un tiempo de 5 minutos), de la misma manera el medio de cultivo en el que se presentó una mayor formación de callos fue el medio MS modificado N°6 (con una diferencia significativa de p>0.005). Concluyendo así que el empleo de hormonas de regulación como AIA, 2,4D y 6-BAP en los medios de cultivo, indujo el proceso de la organogénesis indirecta en todas las concentraciones evaluadas, lográndose un 100% de formación en Me6 a concentraciones de 500 uL; 2000 uL 500 uL respectivamente.
- PublicaciónAcceso abiertoDeterminación estadística de la influencia de los parámetros del control de calidad del agua potable de los municipios de Floridablanca, Girón y Bucaramanga(Bucaramanga : Universidad de Santander, 2018, 2018-01-22) González Sáenz, Ana M.; Osorio Márquez, Jorge-Daniel; Orlandoni Merli, GiampaoloDrinking water is essential for human beings and due to this, the companies that supply this resource must maintain control over the physicochemical and microbiological characteristics after a purification process. The results of the physicochemical and microbiological analyzes for the case of Colombia, are compared with the maximum permissible limits established in resolution 2115 of 2007, which additionally with the calculation of the drinking water quality risk index (IRCA) allows to establish a criterion of quality of the treated water. This work intends to determine, through statistical tools, the influence of the parameters analyzed in drinking water and its correlation with the IRCA; having as main purpose, to know the dynamics of these variables. According to the descriptive statistical analysis, it was found that for the three municipalities the chlorine residual and pH parameters showed a low coefficient of variation (<10%) and hardness, in Bucaramanga and Floridablanca, while the parameters color, heterotrophic count, aluminum and iron had a high coefficient of variation (>30%). On the other hand, in the linear regression model, the variables chlorides, sulphates, aluminum hardness and alkalinity had a positive correlation for the three municipalities with a correlation coefficient between 0.50 and 0.90. The principal component analysis significantly reduced the dimensionality of the parameters evaluated in the study, indicating that turbidity is the major correlation parameter of the IRCA. The model failed to determine significantly the quality of the treated water. The goodness of fit only explained 48% (Bucaramanga) and 52% (Floridablanca and Girón) with respect to the calculation of the IRCA, which requires implementing or completing the model with other types of data analysis techniques to determine a greater relationship with IRCA and its purpose.
- PublicaciónAcceso abiertoConstrucción de un biofiltro para la remoción biológica de sulfuro de hidrógeno en aire utilizando residuos de Moringa oleífera(Bucaramanga : Universidad de Santander, 2018, 2018-05-24) Mora Toloza, Brisley Yuliana; Zafra, German; Chacín Zambrano, Christian-AndreiHydrogen sulfide (H2S) is a colorless, flammable, toxic gas with a characteristic "rotten egg" odor, produced by the activation and reduction of sulfate to sulfides. Currently, there are several chemical and biological methodologies, used for the elimination of this compound. However, the chemical or physical strategies typically transform the contaminant to a more toxic compounds and have a low removal efficiency. This is why there is a need of optimizing the biological technologies optimized for the treatment or elimination of H2S. Therefore, the objective of the present study was to develop an airlift biphasic biofilter using Moringa oleifera residues as an immobilizing material, under different retention times and contaminant concentrations. The biofilter was efficient to remove H2S in contaminated air. The biofilter removal efficiency was not affected by the inlet H2S concentrations; however, a direct relation was found between the retention time and gas removal with significant differences between 0 and 15 minutes as well as 0 and 30 minutes, but not between 15 and 30 minutes retention. In conclusion, it was possible to build an efficient biofilter for the removal of H2S in air. The residues of Moringa oleifera husks have shown on their own a retention and adsorption effect on the H2S present in the contaminated air; likewise, the biofilter built with Moringa oleifera residues was efficient to eliminate up to 65% of the H2S present in polluted air currents.

To contribute to this purpose, there are analysis techniques that allow to evaluate the quality of the seeds for the company, which are of interest both for the seed industry and the institutions responsible for the certification, which determine the value of the seeds for profit. of the farmer. In this study, the morphological characteristics and the levels of natural fungal infection in the germination of seeds of Sacha inchi (Plukenetia Volubilis) from different regions of Colombia were evaluated through statistical methods. Initially, the establishment of the morphological variability of the sacha inchi seeds selected from the regions of Santander, Valle, Antioquia, Boyacá and Cesar was determined, determining the size, texture, color, weight and shape, from which a classification of 3 groups defined with 93% correct data through discriminant analysis. Subsequently, the percentage of germination, level of cultivable fungal infection was determined. The germination results were 92% in a global average of the regions and an infection percentage of 95% when the Macrophomina genus had a higher frequency of appearance in all the regions studied. Then, comparisons were made to the germinative capacity according to the region of origin and it was evidenced that the Boyacá region was the slowest to germinate compared to the Santander region that achieved its germination in the first week of the study. It was possible to generate a contribution to the improvement of quality in the selection of sacha seeds according to a characteristic taking into account the different regions evaluated, being a basis for future research related to the morphology and health of the seeds.
- PublicaciónAcceso abiertoEvaluación de la capacidad Fotosintética de Heliconiapsittacorum (L.f.) asociada a Rhizophagus irregularis y Asperguillus niger en suelos impactados por cadmio(Bucaramanga : Universidad de Santander, 2018, 2018-06-28) Aguillón Almario, Diana Marcela; García Castro, Stefanía; Guerra-Sierra, Beatriz ElenaThe presence of an excessive amount of cadmium in the soil causes various symptomatology in plants, such as reduced growth especially root growth alterations in mineral nutrition and carbohydrate metabolism and can therefore strongly reduce biomass production. This reduction of the biomass due to the toxicity of cadmium (Cd) could be one of the most important consequences in the inhibition of chlorophyll synthesis and in the decrease of the photosynthetic rate of the plants. That is why the association of arbuscular mycorrhizal fungi with the plant confers nutritional benefits and resistance to different stress factors, both biotic and abiotic. The objective of the present was to study the capacity of photosynthetic production in plants of Heliconia psittacorum in association with mycorrhizal fungi under light and shade conditions, in three soils naturally impacted by cadmium (0.96, 1.075, 18.83 ppm) present in the native soils of San Vicente de Chucuri. In this work the response variables were evaluated such as mycorrhizal symbiosis, total chlorophyll amount, physiological differences in leaf growth and stem length and quantification of root biomass. The factors evaluated allowed to show that each of the treatments for each factor such as concentration of cadmium, inoculated and light source is correlated and present significant differences among themselves. In conclusion, the use of mycorrhizae in soil contaminated with cadmium increases the photosynthetic capacity and favors the growth and development of the plant as well as its root biomass, since this symbiosis retains help to retain cadmium, preventing it from being disturbed to other parts of the soil. The plant, likewise it is important to mention that the photosynthetic rate was not affected by the concentration of cadmium present in the soil.
- PublicaciónAcceso abiertoIdentificación y evaluación de la actividad metabólica de microorganismos contaminantes representativos de la etapa de elaboración de azúcar.(Bucaramanga : Universidad de Santander, 2018, 2018-06-29) Guerrero Gómez, Lucila Selene; Daza Merchán, Zunny Tatiana; Zafra, GermanIn the process of making sugar, where the conditions of temperature and osmotic pressure are not favorable for the development of microorganisms, there has been an increase in the concentration of microbial metabolites that influence the physical-chemical characteristics of the materials and affect the process . The objective of the present investigation was to characterize microorganisms isolated from the process of the elaboration stage and to identify their possible metabolic activity in substrates and conditions of the process. Isolations were made in three pilot mills and microorganisms were identified by gene sequencing. BAL lactic acid bacteria belonging to genera Lactobacillus, Entrococcus, Weisella and Bacillus were identified; yeasts of genera Meyerozyma, Pichia, Candida, Trichosporon, Rhodotorula, Saccharomyces and Debaromyceni; thermophilic bacteria of the genus Bacillus, and lactic acid bacteria producing exopolysaccharide (BALPE) from genera such as Leuconostoc, Lactobacillus and Bacillus. In order to know the possible effect of the microorganisms in clear juice and molasses, deterioration tests were performed by inoculating microorganisms representative of each microbial group and evaluating the behavior at 30 ° C and 60 ° C. The statistical analysis of the response variables of sucrose consumption and production of organic acids was carried out. It was observed that the greatest effect in terms of sucrose inversion is caused by the yeasts and in terms of production of metabolites are the BAL that produce the highest amount of metabolites in juices at room temperature and thermophilic bacteria at temperatures of 60 ° C . We identified genes involved in the production of exopolysaccharides such as the dextransucrase gene in Leuconostoc pseudomesenteroides and the levansucrase gene in Lactobacillus and Bacillus genera. This work made it possible to demonstrate that under conditions of high temperature and high concentration of sugars characteristic of the production processes, thermophilic bacteria are the main responsible for the increase of metabolites in this area.

The enzyme responsible for the reduction of sulfate disassimilation is encoded by different genes, including the most studied gene dsrA, used for the detection and quantification of these bacteria. In this work, was standardized the polymerase chain reaction (PCR) technique for the detection of the DsrA gene, using new oligonucleotides previously designed by the young investigator Karen Angarita. For this, an analysis was made "in silico" of the control oligonucleotides and the oligonucleotides with potential to amplify the gene of interest, three different polymerases were evaluated, and the respective detection limits were determined. The amplified products were analyzed using agarose gels at 1.5%. The genomic DNA of Desulfovibrio vulgaris (ATCC 29579) was used as a positive control. The results show that the oligonucleotides designed have the potential to be used to detect sulfate reducing bacteria, since they showed no DNA amplification of other possible bacteria present in samples of interest in the industry Oil and with a detection limit of 1×10−5 ng of Desulfovibrio vulgaris DNA.

The execution of this project is aimed at developing in the municipality of La Belleza, Santander, in order to venture into the wine market and contribute to the development of this region, the company Passiflora has the entire production area and infrastructure necessary for the elaboration of the artisanal drink. This work was carried out in conjunction with the financial management area of the University of Santander UDES responsible for evaluating the financial aspects required by the project, on the other hand, the development of winemaking was led by the industrial microbiology staff helping to give distinction and product quality, taking into account that it will be totally handmade, the different equipment will be available to maintain control in the process of winemaking and this ensure that it complies with the legal specifications for marketing.
- PublicaciónAcceso abiertoCaracterización de la diversidad microbiana cultivable, asociada al residuo lignocelulósico del proceso de beneficio de la hoja fique (Furcraea Andina) en Mogotes, Santander.(Bucaramanga : Universidad de Santander, 2018, 2018-07-26) Vargas Gutiérrez, María Paula; Pinilla Corzo, RigobertoThe purpose of the present study was to contribute to the recognition of the diversity of microorganisms present in the residue lignocellulosic from the process of benefit of the sisal leaf Furcraea andina, the samples were taken at the experimental farm Jose Exelino Moreno, located in the municipality of Mogotes, Santander. The isolation of the culturable microorganisms was given through the use of specific culture media, after performing serial dilutions up to 10-6, later the microbial groups associated whit this residue were established by means of morphological keys and biochemical tests and finally the abundance of microorganisms were determined for each of the samples collected. We obtained a total of 55 isolates correpondian to 36 bacteria, 6 fungi and 13 yeast. In the macroscopic and microscopic characterization, 20 bacteria were selected 70% of the bacteria were Gram negatives and 30% Gram positive. Subsequently whit biochemical tests it could be deduced that there may be the presence of the family Enterobacteriaceae and of the genera Pseudomonas sp. and Bacillus sp. In the same way for the fungi, 7 isolates were selected, of which according to the technique of micro-culture and the taxonomic keys, one finds in greater proportion the genus Penicillium sp.. Finally, we selected 8 isolates of yeast, which according to the results obtained in the API 20 AUX, is the presence of two microorganisms of industrial interest Candida famata y Rhodotorula glutinis, the other yeasts according to the literature are reported as pathogenic for humans. Finally, the isoletes characterized were retained for the purpose of maintaining this biological material in a viable and stable state by method tube whit angle agar in mineral oil.

The purpose of this degree work is to propose a service through a business model, taking advantage of a market that improves the image and profit margin of the company. First, the market of Bucaramanga and its metropolitan area was analyzed, and then a technical analysis was built based on the requirements of current legal regulations and technical bibliography. Based on the previous analysis, financial assumptions were projected and a model was finally constructed. Of business, using the Canvas model. The analysis of supply and demand showed that the demand is unsatisfied despite the fact that physicochemical analysis is offered in Bucaramanga and the metropolitan area, the competition does not have technical accreditation of test methods under ISO 17025. The technical and financial analysis determined that if it is viable to implement the business unit, the initial technical investment is 14,758,780 COP, the capacity of the area in disuse is between 30 and 60 monthly samples, the TIR and VNA indicators are greater than 0, which is economically viable, the pay-back shows that the investment recovers in 3 years. The business model helped to reflect how the company operates, its gears and generate the value proposition that will be a fast service, accredited under ISO 17025-2017, in addition to an unconditional accompaniment with the client and his company.
- PublicaciónAcceso abiertoEvaluación de la producción de flavonoides a partir de orégano (origanum vulgare) mediante la técnica de suspensiones celulares(Bucaramanga : Universidad de Santander, 2018, 2018-11-12) Mantilla Arias, Yulieth A.; Chacín Zambrano, Christian-Andrei; Rodríguez González, Leidy-GabrielaEl cultivo de células y tejidos in vitro en suspensión se convierte en una alternativa para la síntesis y obtención de metabolitos secundarios de interés industrial a partir de las plantas, dentro de estos metabolitos podemos encontrar los flavonoides los cuales son moléculas antioxidantes, eliminadores de radicales libres, antiinflamatorios empleados en la industria farmacéutica como cosméticos y medicamentos. Diferentes estrategias in vitro han sido desarrolladas con el fin de aumentar el contenido de metabolitos secundarios como el uso de biorreactores. Los cuales, generan condiciones ideales para procesos de producción. El objetivo de esta investigación producir flavonoides a partir de Origanum Vulgare (Orégano) por medio de suspensiones celulares en biorreactor airlift., como resultado realizaron pruebas colorimétricas como el ensayo de Shinoda, Álcalis, entre otros con resultado positivo, mostrando la posible presencia de flavonas, calconas, flavonoles y flavonoides, además se obtuvo un máximo de producción de flavonoides de 22,587,280 g de catequina por ml de extracto a partir de callos de Origanum vulgare en un periodo de 10 días a 18 ℃. Se evidenció que el oxígeno es una variable importante incidencia el proceso, pues se obtiene una ganancia de 5 días para la producción de flavonoides y existen diferencias significativas (p<0,05) con respecto a la ausencia de aireación en el sistema; y la temperatura, así como la variable tiempo, se convierte en inductores de crecimiento y producción de flavonoides. De esta manera las aplicaciones de nuevas técnicas biotecnológicas generan nuevas posibilidades de desarrollo sostenible y contribuyen con la preservación del medio ambiente.
- PublicaciónAcceso abiertoCaracterización de nuevas proteínas Cry11 obtenidas por modelos heurísticos computacionales(Bucaramanga : Universidad de Santander, 2018, 2018-11-23) Abaunza Villamizar, Sebastián Mauricio; Suárez Barrera, Miguel OrlandoBacillus thuringiensis (Bt) is a Gram positive bacterium with parasporal inclusion bodies, also called δ-endotoxins, within these is the Cry protein that is known for its toxic activity against multiple orders of insects and nematodes, the vector-borne diseases A. aegypti, it transmites several important diseases in public health such as Dengue, Zika Chikungunya, among others. Cry11 has been recognized for its high specificity against A. aegypti. However, the emergence of resistance by insects that are the object of study, have resulted in the application of various strategies, for the improvement of proteins including site-directed mutagenesis and DNA shuffling. The use of biocomputational tools has expanded the spectrum in the development of molecules with greater potential, however, these strategies have not been described in the development of enhanced mutants of Cry. This work is based on the use of software HIDDEN 1.0 where libraries of in silico variants were obtained from Cry11Aa. Heu2, Heu3 and Heu4 variants were selected, and validated in vitro, they were synthesized, cloned into PSV2 and were expressed in E.coli DE3BL21 and B. thuringiensis BMB171, their electrophoretic profiles were analyzed in SDS-Page and their toxicity activity was evaluated by thick tests with A. aegypti. Parallel to this a structural analysis with matrix of MatGat, Bioedit and structural comparisons were contrasted with the literature. It was found that the variants presented an upper identity more than 97% with Cry11Aa, the changes mostly belonged to domain II, and their lethality percentages were less than 5%, which suggests that the amino acid changes in the important regions of the three domains are involved in the toxicity activity demonstrated by the proteins.

Cry11 is a specific toxin against the vector A. aegypti, which is responsible for the transmission of dengue, zika and chikungunya; however, its mode of action and structure-function characteristics have not yet been fully elucidated. The research group of Molecular Biology and Biotechnology of the UDES, has a library obtained by shuffling the DNA of cry11 genes, highlighting the variant 8Cry11, which is 6 times more toxic than Cry11Aa and 3.8 more than Cry11Bb. Molecular Docking studies showed that positions 553 and 556 of this protein are relevant in the interaction with the cadherin receptor, to corroborate this information, site-directed mutagenesis was performed to reverse the aforementioned mutations, obtaining the variants 8Cry11L553F, 8Cry11L556W, and 8Cry11L553F-L556W. In this work, the toxic activity of mutants 8Cry11L553F, 8Cry11L556W, and 8Cry11L553F-L556W was determined, as well as an approximation of protein analysis both in silico and in vitro through SDS-PAGE. To achieve this, the ideal conditions for the production of δ-endotoxin (Cry11Aa) were standardized, finding a relationship between glucose concentrations (15g / L) and sources of organic and inorganic nitrogen in a ratio of 3: 7; the production of protoxin (~ 100 kDa) and toxin (32 and 34 kDa) was corroborated by SDS page. To determine the mean lethal concentration in comparison with the mutant 8Cry11 and the parental Cry11Aa, the toxicity of the mutants was evaluated, against first stage larvae of A. aegypti. The results showed loss of toxicity for the variants under study, which indicated that substituted amino acids in domain III were strongly possible involved in the loss of toxicity due to structural features.
