Determinación de la Actividad Antimicrobiana de los Péptidos Ib-M Frente a Staphylococcus aureus ATCC 25923 y Candida albicans ATCC 10231

Los péptidos antimicrobianos (PAM) se han convertido en una alternativa promisoria para el tratamiento de enfermedades infecciosas a nivel mundial, teniendo en cuenta la generación de resistencia antimicrobiana (RAM) por microorganismos como Staphylococcus aureus y Candida albicans. Los PAM son en el objetivo principal de múltiples investigadores, teniendo en cuenta que son moléculas efectoras de la inmunidad innata, la mayoría poseen carga positiva, actividad anfipática, antimicrobiana, poseen baja citotoxicidad y se ha logrado demostrar que la gran mayoría interaccionan con la membrana celular de los microorganismos y con ello se perturba la generación de RAM. La familia de péptidos Ib-M, han sido utilizados desde 2014 por Flórez y Colaboradores en diversos estudios en donde se ha demostrado que poseen alta hidrofobicidad, un bajo efecto hemolítico y citotóxico y una carga de +6. Adicionalmente, se ha demostrado que son activos frente a cepas de Escherichia coli, O157:H7 y K-12 No patógena, en donde radica la importancia de verificar la actividad antimicrobiana frente a otros microorganismos. La metodología utilizada para la determinación de la CMI fue la técnica Microdilución descrita por la CLSI de 2018 en los protocolos M07-A9 para Staphylococcus aureus y M27-A3 para Candida albicans, en donde se construyen cinéticas de crecimiento basadas en valores de absorbancia. Adicionalmente se verifico la reproducibilidad del método bajo la técnica Difusión en disco, en donde el péptido no difunde correctamente en el disco por variables no diferenciales. Finalmente se encontró que los péptidos Ib-M, 1, 2 y 6 tienen una CMI de 50 µM frente a S. aureus, en 24 horas de prueba. Por otro lado, se precisa un CMI de 25 µM para el Ib-M (2 y 6) y de 100 µM para el Ib-M1 hasta las 24 horas de prueba, en un tiempo total de incubación de 48 horas.

Antimicrobial peptides (AMP) have become a promising alternative for the treatment of infectious diseases worldwide, considering the generation of antimicrobial resistance by microorganisms such as Staphylococcus aureus and Candida albicans. AMPs are the main objective of multiple researchers, considering that they are effector molecules of innate immunity, most of them have a positive charge, amphipathic, antimicrobial activity, low cytotoxicity and it has been demonstrated that the vast majority interact with the membrane. of microorganisms and thereby disturbs the generation of antimicrobial resistance. The Ib-M family of peptides have been used since 2014 by Flórez and Collaborators in various studies where it has been shown that they have high hydrophobicity, a low hemolytic and cytotoxic effect, and a charge of +6. Additionally, it has been shown that they are active against strains of Escherichia coli, O157:H7 and K-12 Non-pathogenic, where the importance of verifying the antimicrobial activity against other microorganisms lies. The methodology used to determine the MIC was the Microdilution technique described by the 2018 CLSI in protocols M07-A9 for Staphylococcus aureus and M27-A3 for Candida albicans, where growth kinetics are constructed based on absorbance values. Additionally, the reproducibility of the method was verified under the disk diffusion technique, where the peptide does not diffuse correctly in the disk due to non-differential variables. Finally, peptides Ib-M, 1, 2 and 6 were found to have an MIC of 50 µM against S. aureus, in 24 hours of testing. On the other hand, a MIC of 25 µM is required for Ib-M (2 and 6) and 100 µM for Ib-M1 up to 24 hours of testing, in a total incubation time of 48 hours.
