Iron is one of the most abundant elements on Earth and the most abundant metal in the human body. This element is decisive for the development of life because almost all the organisms need iron to perform different biological activities. In humans, the concentration of Fe is regulated by several proteins that contain this element, such as transferrin (TF). TF carries Fe to every cell that needs it and keeps it away from pathogens. These organisms have developed different strategies for obtaining iron. One of them is the expression of specific transferrin receptors located in the plasma membrane. This is the case of Entamoeba histolytica, a microorganism that requires Fe to survive and in which it has been observed that TF binds to its receptor, signal transduction pathways related to its activation are important in proliferation.The study of transferrin receptors and other proteins related to signaling pathways is important because these proteins could be used as therapeutic targets due to their specificity. In this report, both the culture and the obtaining of proteins that present a state of phosphorylation by the use of TF in E.histolytica are described. The expected scope of the present work is to contribute to the study of the use of the transferin by identifying the proteins that participate in the signaling pathways and that are useful as therapeutic targets.

El hierro es uno de los elementos más abundante en la Tierra y el metal más cuantioso en el cuerpo humano. Este elemento es decisivo para el desarrollo de la vida porque casi todos los organismos necesitan hierro para realizar diferentes actividades biológicas. En los humanos la concentración de Fe está regulada a través de varias proteínas que contienen este elemento, como la transferrina (TF). La TF transporta Fe a cada célula que lo necesita y lo mantiene alejado de los patógenos. Estos organismos han desarrollado diferentes estrategias para la obtención de hierro. Uno de ellos es la expresión de receptores de transferrina específicos localizados en la membrana plasmática. Es el caso de Entamoeba histolytica, microrganismo que requiere de Fe para sobrevivir y en el que se ha observado que una vez que la TF se une a su receptor las vías de transducción de señales relacionadas con la activación de este tienen importancia en la proliferación. El estudio de los receptores de transferrina y otras proteínas relacionadas con las vías de señalización es importante porque estas proteínas podrían usarse como dianas terapéuticas debido a su especificidad. En este informe, se describe tanto el cultivo como la obtención de proteínas que presentan un estado de fosforilación por la utilización de la TF en E.histolytica. El alcance esperado del presente trabajo es contribuir al estudio del uso de la transferina mediante la identificación de las proteínas que participan en las vías de señalización y que son útiles como objetivos terapéuticos.
