Many species of Acanthamoeba spp. they are responsible for fatal diseases for humans such as: meningoencephalitis and keratitis, so the study of their endosymbiotic interaction is representative. The purpose of the study was to establish the endosymbiosis between Acanthamoeba spp. and Cryptococcus spp. in environmental samples from the city of Cúcuta, Norte de Santander. A descriptive study was carried out, collecting a total of 132 samples, 120 of soils from four parks in the city of Cúcuta, Norte de Santander: Colón Park, Santander Park, San Eduardo Park, Erasmo Meoz University Hospital Park and 12 water samples taken from the Pamplonita River and the Bogotá Wastewater Canal. The culture was carried out on non-nutritive agar plates previously seeded with inactive Escherichia coli ATCC 25922. Acanthamoeba spp. it was isolated from 83 (70%) of the soil samples and 12 (100%) of the water samples. Also, the identification of Cryptococcus spp. as an endosymbiont, it was present in 7 (5.8%) of the strains samples of Acanthamoeba spp.

Studies carried out by the Group of works of the UDES, have allowed the conformation of a bank of strains constituted by clinical and environmental samples of Cryptococcus, it is important to know that there are references on the use of the Galleria mellonella model for the study of the pathogenesis of Cryptococcus neoformans highlighting the advantages of this simple model, unlike the mammals models where their manipulation is more complicated and they are expensive. Therefore, the general objective of the project was the evaluation of the pathogenicity of Cryptococcus neoformans serotype VNI in Galleria mellonella. To this end, two control strains of Cryptococcus (H99 and JEC21) were used, which correspond to the variety grubbi and the variety neoformans, respectively, which, like the strains studied, were incubated and subsequently inoculated in the invertebrate model G. mellonella, Pathogenicity modulation was evaluated by survival curves by the Kaplan mejer statistical method, where the days of death of the larvae caused by each of the clinical and environmental isolates were observed. Additionally, the capsular size of the Cryptococcus studied was evaluated, because the capsule is the main factor of virulence. The capsule was measured in fresh preparations 15 with Chinese ink before and after infection, in the same way the adaptive changes of Cryptococcus neoformans were evaluated, submitting them to an oxidative stress with NaOH, HCl and Menadione. This work made important contributions on the pathogenicity characteristics of both clinical and environmental Cryptococcus strains of the region, thus contributing to the development of studies of Cryptococcosis.
